This is a step-by-step hand holding, descriptive, illustrated picture guide to make Escherichia coli (E. Coli) grow with a green fluorescent protein (GFP), so it glows green when hit with ultraviolet light (UV). The goal is to help folks gain confidence in basic lab processes and equipment while also getting descriptive explanations about the biological interactions happening at each step.

Time / Scheduling

Start → End: 2 Days (E Coli. growth happens overnight)

Focused Lab Work: 4-5 Hours

Stage Time/Days Schedule
Stage 1: Plate Making 2 1/2 hours Friday (Evening)
Stage 2: Plating E. Coli 2 hours
Stage 3: Incubation Overnight (12-16 hrs)
Stage 4: Pick Up 15 Minutes Saturday (Morning)

Steps

  1. Check Materials
  2. Clean & Sterilize Glassware
  3. Prepare Plates (aka Petrie Dishes) with LB Media (Food for Bacteria to Grow On)
  4. Transform (aka Insert) GFP Plasmids into Thawed Out E. Coli Cells
  5. Plate E. Coli Cells (That Now Also Contain GFP Plasmids)
  6. Incubate Plates (Runs Overnight)
  7. Evaluate Results (Next Day)
